and the activated signalling pathways investigated. To ensure equivalent activation of downstream pathways by each ligand, phosphorylated-smad2 levels were assayed by Western blot. Up to 100-fold higher concentrations of Xnr1 than ActivinA were required to induce equivalent levels of phosphorylation of smad2. The expression of well-known downstream targets of Activin and Xnr1 was assayed by quantitative PCR to confirm appropriate pathway activation. Affymetrix Microarrays were then performed at three developmental timepoints to identify highly differentially regulated novel genes, which were then validated by examining their expression patterns in gastrulation stage embryos. Initial experiments to overexpress and knockdown these genes are currently being performed.

Alsin is a Rab5 and Rac1 guanine nucleotide exchange factor.
ALS2 is the gene mutated in a recessive juvenile form of amyotrophic lateral sclerosis (ALS2). ALS2 encodes a large protein termed alsin, which contains a number of predicted cell signaling and protein trafficking sequence motifs. To gain insight into the overall function of alsin and to begin to evaluate its role in motor neuron maintenance, we examined the subcellular localization of alsin an...

ALS2/alsin is a member of guanine nucleotide exchange factors for the small GTPase Rab5 (Rab5GEFs), which act as modulators in endocytic pathway. Loss-of-function mutations in human ALS2 account for a number of juvenile recessive motor neuron diseases (MNDs). However, the normal physiological role of ALS2 in vivo and the molecular mechanisms underlying motor dysfunction are still unknown. To ad...

Primary lateral sclerosis (PLS) is a rare progressive paralytic disorder that results from dysfunction of the upper motoneurons. Although PLS is a sporadic disorder of adult middle age, it has also been described in children as juvenile PLS or JPLS. The causative gene for JPLS was found to be ALS2, which is also responsible for a recessive form of amyotrophic lateral sclerosis, for infantile on...
متن کاملRab5 and Alsin regulate stress-activated cytoprotective signaling on mitochondria
Mitochondrial stress response is essential for cell survival, and damaged mitochondria are a hallmark of neurodegenerative diseases. Thus, it is fundamental to understand how mitochondria relay information within the cell. Here, by investigating mitochondrial-endosomal contact sites we made the surprising observation that the small GTPase Rab5 translocates from early endosomes to mitochondria u...
متن کاملDrosophila Strip serves as a platform for early endosome organization during axon elongation
Early endosomes are essential for regulating cell signalling and controlling the amount of cell surface molecules during neuronal morphogenesis. Early endosomes undergo retrograde transport (clustering) before their homotypic fusion. Small GTPase Rab5 is known to promote early endosomal fusion, but the mechanism linking the transport/clustering with Rab5 activity is unclear. Here we show that D...
